A rice tubulin tyrosine ligase‐like 12 protein affects the dynamic and orientation of microtubules

The detyrosination/retyrosination cycle is the most common post-translational modification of α-tubulin. Removal conserved C-terminal tyrosine α-tubulin by a still elusive tubulin carboxypeptidase, and religation this ligase (TTL), are probably to all eukaryotes. Interestingly, for plants, only candidates qualifying as potential TTL homologs ligase-like 12 proteins. To get insight into biological functions these homologs, we cloned rice TTL-like protein (OsTTLL12) generated overexpression OsTTLL12-RFP lines in both tobacco BY-2 cells. We found, unexpectedly, that increased relative abundance detyrosinated coleoptile seminal root, correlated with more stable microtubules. This was independent respective orientation cortical microtubule, followed correspondingly changing growth coleoptiles roots. A perturbed organization phragmoplast microtubules disoriented cell walls were further characteristics phenotype. Thus, elevated detyrosination consequence OsTTLL12 affects structural dynamic features microtubules, changes axiality plate deposition and, consequently, plant growth.
